This product is manufactured by Vertex Pharmaceuticals and is a combination of 3 ingredients, tezacaftor, vanzacaftor, and deutivacaftor.

Why is it prescribed?
Alyftrek™ is indicated for the treatment of cystic fibrosis (CF) in patients 6 year of age and older who have certain mutations in the cystic fibrosis transmembrane conductance regulator (CFTR) gene.
- This medication should always be taken just before or just after fat-containing food. Avoid foods and drinks containing grapefruit.
- Take each dose once daily at approximately the same time.
- Swallow the tablets whole. Do NOT chew, break or dissolve the tablets.
- If you miss a dose and it has been within 6 hours of the time you usually take it, take it as soon as possible.
- If you miss a dose and it has been more than 6 hours since the time you usually take it, skip the missed dose and take the next dose at your regularly scheduled time. Do NOT double your dose.
- Store at or below 30°C.

Dosage for individuals 6 year and older:
- < 40 kg: 3 tablets of vanzacaftor 4 mg/ tezacaftor 20 mg/ deutivacaftor 50 mg once daily
- ≥ 40 kg: 2 tablets of vanzacaftor 10 mg/ tezacaftor 50 mg/ deutivacaftor 125 mg once daily
Various mutations of the cystic fibrosis gene block the "gates" that allow the flow of salt across the cell membranes. This causes mucus to become thick and clog the lungs. Alyftrek™ helps "open the gates" more often allowing more salt to pass through. The thick mucus is reduced and cystic fibrosis symptoms lessen.
Important potential side effects. These are not common but are possible:
- liver problems
- cataracts
- allergic reactions
Contact your doctor if any of these side effects persists or are bothersome:
- headache
- diarrhea
- rash
- This medication may cause liver problems.
- Do not take this medication if you are allergic to any part of it.
Take this medication with caution if you:
- have liver problems
- have kidney problems
The following medications may interact with Alyftrek™:
- antifungal medicines such as ketoconazole, itraconazole, posaconazole, voriconazole, fluconazole
- some antibiotics such as clarithromycin, rifabutin, rifampicin, erythromycin
- some anti-seizure medications such as phenobarbital, carbamazepine, phenytoin
- St. John’s wort
- some medications used after an organ transplantation such as cyclosporine, everolimus, sirolimus, tacrolimus
- efavirenz
- digoxin
- warfarin
- medications that are substrates of the breast cancer resistance protein (BCRP) such as methotrexate and glyburide
- some medications used to treat diabetes such as glimepiride, glipizide
- some medications that lower blood pressure such as verapamil
- products containing grapefruit
